Abstract: Triglycerides constitute the majority of the body`s lipids,and it is these molecules that are generally referred to simply as "fat". The measurement of triglycerides in human serum is very important for clinical use in hospitals. In this thesis, a method was developed for this purpose. The work include analyzer modification, instrumentation adjustment and design of test programs etc.. The method used is enzymatic colorimetric method specifically adaptable to The OptimateTM Automated Fluorometer and Photometer manufactured by Ames Company, USA. In this method, triglycerides were hydrolyzed by lipoproteinlipase to glycerol and fatty acid. Through a series of intermediate reactions, the resulted glycerol was finally changed into a violet coloured compound which can then be determined photometrically. The increase in absorbance is proportional to the concentration of triglycerides in human serum samples. The developed method is proved to be easy to operate. Once started it follows the designed program in the microprocessor. After preparing the reagents, blank, standard and sample, putting them in place and setting the correct volume for reagent and sample, then the tests goes automatically. The method share the common sequence of enzymatic hydrolysis of triglycerides followed by enzymatic determination of the resulting glycerol. It is shown to be accurate and efficient. Through my tests, it correlate well with the method used in the Tampere University Hospital, provided that special attention is paid to the calibration of analyzers. /Kir10
